Screening of individual radiosensitivity using the γ-H2AX assay: differences between high-LET and low-LET radiation

Abstract

This thesis was carried out within the larger framework of the ESA-IBER 3 GYMBRASS project at the Radiobiology Unit of SCK•CEN. This study aims to identify biomarkers such as genes, exons, microRNAs and secreted proteins which can be used for biodosimetery with the purpose of tracing radiation exposure as well as predicting individual radiation sensitivity to heavy ions. The aim of the thesis was the optimization of an assay for the detection and quantification of -H2AX foci in peripheral blood mononuclear cells (PMBCs) from healthy donors following exposure to X-ray irradiation. In this regards, screening individuals for -H2AX formation and repair can shed light on the individual response towards ionization radiation. For the GYMBRASS project the optimization of this protocol will be utilized to determine the individual radiosensitivity to low- and high- linear energy radiation transfer radiation of each individual donor, both for radiation-induced DSB formation as well as for DNA repair kinetics. Finally these data will be incorporated with those of gene and cytokine expression from the same donors with the aim of identifying molecular biomarkers for individual radiosensitivity.
